Abstract |
The effect of TGF-beta1 on the growth of Y-79 retinoblastoma cells was examined. Y-79 retinoblastoma cells treated with TGF-beta1[0.5 ng/ml, 1.0ng/ml] were cultured and their growth curve was compared to the untreated cultured cells. Phase contrast microscopy, transmission electron microscopy and flow cytometer were also performed. Administration of TGF-beta1 significantly inhibited growth of Y-79 cells[p<0.05]. Transmission electron microscopic examination of those cells showed nuclear condensation, fragmentation and cytoplasmic blebbing, suggesting apoptosis as an underlying mechanism. And apoptosis was quantified by flow cytometric analysis. TGF-beta1 significantly induced apoptosis in Y-79 cells[p<0.05]. The histogram of TGF-beta1 treated Y-79 cells showed an increase in G2/M populations and decrease in G1 cell populations. This study suggested that TGF-beta1 induced apoptosis in Y-79 retinoblastoma cells, and the most plausible mechanism of growth inhibition and this function might be related to G2/M rather than G1 of the cell cycle related to Rb protein. |
